Introduction

Phospho peptides are essential tools to study phosphorylation by protein kinases in many eukaryotic cellular processes.  The specificity of the post-translational phosphorylation is critical in this kind of research.

Multiple Phospho Peptides

Since there are three possible amino acids for phosphorylation; Tyr, Thr, and Ser, it is essential to prepare peptides with a phosphate group at a specific site, as well as peptides with more than one phosphate group at defined amino acids.

Princeton BioMolecules has developed an unique service for custom synthesis of single and multiple phosphorylated peptides with following specific characters:

1. site specific on Tyr, Thr, and/or Ser (examples 1, 2, 3, 4 and 5)

2. additional groups such as biotin can be introduced at specific sites (examples 3 and 5)

3. multiple phospho-residues on a single peptide (examples 6 and 7)

Examples

The following are some examples of multiple phosphorylated peptides synthesized at Princeton BioMolecules:

1. [Ac]- Gly-pTyr-Met-Asp-Met-Ser-Lys-Asp-Glu-Ser-Ile-pTyr-Val-Pro-Met-Leu-[NH2]

2. [H]- Tyr-Asp-Met-Thr-Thr-pSer-Gly-pSer-Gly-Ser-Gly-Leu-Pro-Leu-Leu-Cys -[OH]

3. [H]- Tyr-Asp-Met-Thr-Thr-pSer-Gly-pSer-Gly-Ser-Gly-Leu-Pro-Leu-Leu-Cys(Biotin) -[OH]

4. [H]- Lys-Ala-Ile-Glu-Thr-Asp-Lys-Glu-pTyr-pTyr-Thr-Val-Lys-Asp- [OH]

5. [Biotin]- Lys-Ala-Ile-Glu-Thr-Asp-Lys-Glu-pTyr-pTyr-Thr-Val-Lys-Asp- [OH]

6. [H]- Lys-Glu-Leu-Leu-Pro-pThr-Pro-Pro-Leu-pSer-Pro-Ser-Arg-Arg-Ser -OH]

7. [H]- Tyr-Asp-Met-pThr-pThr-pSer-Gly-pSer-Gly-pSer-Gly-Leu-Pro-Leu-Leu-Cys -[OH]
